GAP-43 or growth associated protein 43 (also referred to as neuromodulin, B-50, p57 and F1) is a nervous system-specific phosphoprotein and a major component of motile growth cones which form the leading edge of the elongating axon. The membrane skeleton of the growth cone is responsible for transducing extracellular guidance cues into morphological alterations mediated by the polymerization/de-polymerization of actin resulting in directional changes in axon growth. GAP-43 is closely associated with the membrane skeleton and has been implicated in regulating interactions between cytoskeletal components and the plasma membrane. Interestingly, axonal transport of GAP-43 occurs only in neurons that are extending axons during development or regeneration after injury; however, this transport does not occur in axons that do not normally regenerate such as those in the mammalian central nervous system. |
