The protein contains a signal sequence, a prodomain with both a potential cysteine switch and a furin cleavage site, a metalloproteinase domain with a conserved zinc-binding motif and a 'met turn,' a disintegrin-like domain, and a spacer region between a thrombospondin type 1 motif and thrombospondin submotif. At least four transcript variants have been reported. ADAMTS5 has been implicated in inflammatory joint diseases. ADAMTS5 expression has been documented in a range of normal human tissues.
