E2F1 is a transcription activator that binds DNA cooperatively with the differentiation regulated transcription factor proteins (DP). Together they bind to the promoter region of a number of genes whose products are involved in cell cycle regulation or DNA replication. E2F1 also binds to the retinoblastoma protein (RB1), which functions in the control of cell-cycle progression from G1 to S phase. E2F1 can also mediate p53-dependent and independent apoptosis, and phosphorylation of E2F1 at serine 364 results in its stabilisation and apoptosis induction.
