The rat homolog was first identified in a screen of a rat brain cDNA library using a domain of the alpha subunit of the heterotetrameric AP2 complex as bait. The AP2 complex is one of two major coat proteins of endocytic vesicles that function in receptor-mediated endocytosis and the recycling of synaptic vesicles. AAK1 is responsible for phosphorylating the mu subunit of AP2 to promote clathrin-mediated endocytosis.
