Specificity | This antibody is specific for I?¨¬B?¡¾ and is phosphorylated at the position of serines 32 and 36. The antibody was evaluated by ELISA, dot blot and Western blot. It only recognizes the phosphorylated serines 32 and 36 of I?¨¬B, not other phosphorylated sites or non-phosphorylated peptides. By immunoblot assay, this antibody recognizes ~40 kDa band in the TNF-treated Hela or 3T3 whole cell lysates. It regulates the expression of a number of proteins involved in the regulation of cell survival and immune responses (2). The biological activity of NF-?¨¬B is tightly controlled by its inhibitor protein, I?¨¬B, which binds to NF-?¨¬B and sequesters it in the cytoplasm. There are three isoforms of the I?¨¬B: I?¨¬B?¡¾, -?©÷ and u¡°u. I?¨¬B?¡¾, the well-characterized member of the I?¨¬B family contains both nuclear localization sequence and nuclear export signal that render to I?¨¬B?¡¾ the activity to rapidly shuttle between the nucleus and the cytoplasm. The I?¨¬B?¡¾ protein binds to NF-?¨¬B dimmers, masking their nuclear localization sequences and causing their cytoplasmic retention.
