Background | One of the epithelial cell adhesion molecules, E-Cadherin, plays an important role in the formation of cell-cell contacts in epithelia irrespective their origin form ecto-, meso- or endodermal tissue. This early adhesion event between epithelial cells is a prerequisite for the assembly of all elements of the junctional complex. Furthermore, E-Cadherin plays a crucial role in the maintenance of the epithelial junctional complex and is as such an important molecule in maintaining epithelial integrity. Over 90% of the malignant tumors are carcinomas. One of the prerequisites for the release of carcinoma cells from the primary site might be a defect in intercellular adhesion mediated by the absence of E-Cadherin expression. Therefore, the expression of E-Cadherin might be an important parameter for the determination of the invasive potential of epithelial neoplasms, and for the transition of a benign to a malignant neoplasm. |

Application Note | WB: Use at a dilution of 1:50-1:500. ICC: Use at an assay dependent concentration. IHC-Fr: Use at an assay dependent concentration. IHC-P: Use at an assay dependent dilution. Permeabilization in 0.2% Triton X-100 and pronase digestion is required. Recommended range is 1:25 - 1:100 for immunohistochemistry with avidin-biotinylated horseradish peroxidase complex (ABC) as detection reagent. In frozen and paraffin sections, for dilution use a PBS buffer containing 0.1mM CaCl2 and 0.1mM MgCl2. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |
Specificity | Recognises human E-cadherin, an epithelial cell adhesion molecule located on the lateral border of epithelial cells. In blots, the antibody reacts with the 80 kD and 120 kD bands of E-cadherin.In IHC the reaction is confined to the lateral borders of the epithelial cells, or more intensely in the junctional complex in the small intestine. |
