Background | The two major cytoskeletal proteins implicated in cell motility are actin and myosin. Actin and myosin are constituents of many cell types and are involved in a myriad of cellular processes including locomotion, secretion, cytoplasmic streaming, phagocytosis and cytokinesis. Although actin is one of the most conserved eukaryotic proteins, it is expressed in mammals and birds as at least six isoforms characterized by electrophoresis and amino acid sequence analysis. Four of them represent the differentiation markers of muscle tissues and two are found in practically all cells. |

Application Note | IF: Use at an assay dependent dilution. IHC-P: Use at a concentration of 1 - 2 ¥ìg/ml for 30 min at RT. No special pretreatment is required for immunohistochemical staining of formalin-fixed tissues. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |
Specificity | Reacts with alpha- smooth muscle as well as alpha-skeletal and alpha-cardiac (sarcomeric) isoform of actin. Reacts with tumors arising from smooth muscle (leiomyosarcomas) as well as skeletal muscle (rhabdomyosarcomas). |
Species Cross Reactivity | Cross-reacts with Human, Mouse, Rat, Chicken, Dog, Horse and Pig.Expected to cross-react with Silk worm (98% identity with immunogen) due to sequence homology. Not yet tested in other species. |
