Background | α1-antitrypsin is a plasma protein synthesized in the liver. It consists of a single polypeptide chain and has a molecular weight of 51 kDa. A serine protease inhibitor, its primary function is the protection of lung tissue from the action of neutrophil elastase. The average level of ¡¤1-antitrypsin in plasma is1.3g/L. |
Clonality | Polyclonal |
Host | Sheep |
Isotype | IgG |
Immunogen | Human serum α1-antitrypsin purified from plasma and shown to be homogeneous by SDS-PAGE. |
Antigen Species | Human |
Tested Applications | Double immunodiffusion, Immunoelectrophoresis, Radial immunodiffusion, Rocket immunoelectrophoresis. The usefulness of this product in other applications has not been determined. |
Application Note | RID and Rocket IEP: 2¥ìL antiserum/cm= gel vs 5¥ìL 1/2 - 1/50 dilution human serumDouble diffusion: 10¥ìL antiserum vs 5¥ìL serumIEP: 100¥ìL antiserum vs 5¥ìL serum The use of 3% PEG 6000 with 1.2%agarose in a suitable buffer (such as TBE or Tris-barbital pH >8.2) is recommended. Suitability for use in nephelometry, immunohistochemical procedures, enzyme-linked immunosorbentassays and Western blot has not been assessed but use in such assays should not necessarily beexcluded. |
Specificity | This product gives a single arc when tested by IEP and 2D-IEP against human plasma. Identity is confirmed by double diffusion (Ouchterlony) vs human plasma and a known anti-human ¡¤1-antitrypsin. |
Species Cross Reactivity | Cross-reacts with Human. Does not cross-react with Rabbit, Rat, Horse, Mouse, Cow, Dog, Pig, Cat, Guinea pig or Chicken. Not yet tested in other species. |
Target | alpha 1 Antitrypsin |
Cellular Localization | Secreted |
Purification | IgG fraction |
Purification Note | Antiserum is prepared by immunisation of sheep with human ¡¤1-antitrypsin and, if necessary, adsorbed to monospecificity by use of solid-phase adsorbents. An immunoglobulin fraction is then produced. The titre is adjusted so that inter-batch variation is within 10%. The product is 0.2?m filtered. |
Storage Buffer | Glycine buffered saline pH7.4, 0.1% Sodium Azide, 0.1% EACA, 0.01% Benzamidine,1mM EDTA |
Storage Instruction | Keep as concentrated solution, aliquot and store at 4¡ÆC. |
Notes | Slight precipitation can occur on storage which may be removed by centrifugation and should not affectperformance characteristics. |
