Background | α2-Macroglobulin is a plasma protein, mainly synthesized in the liver. It is a tetrameric molecule, with a molecular weight of 720 kDa and usually occurs in serum at 1000-1500 mg/L. Its main function is as a protease inhibitor. Following conformational change, the protease is deposited within a central cavity where it is still active but prevented from further contact with its substrate. Removal of the complex is mediated by the generation of recognition sites, which react with receptors on a variety of cells including macrophages and liver cells. |

Purification Note | Antiserum is prepared by immunisation of sheep with human ¡¤2-macroglobulin and, if necessary, adsorbed to monospecificity by use of solid-phase adsorbents.An immunoglobulin fraction is then produced. The titre is adjusted so that inter-batch variation is within 10%. The product is 0.2?m filtered. |
