Background | Erythroid band 3 protein (AE1; 109270) is critical for the maintenance of the biconcave disc shape of red blood cells, where it binds to ankyrin. The kidney isoform of band 3 protein, kAE1, does not bind to ankyrin. Chen et al. (1998) cloned a mouse cDNA encoding a protein, termed kanadaptin for kidney anion exchanger adaptor protein, that binds to the cytoplasmic domain of kAE1. The 507-amino acid protein sequence is proline-rich but has no strong homologies to other proteins. Northern blot analysis revealed that in mouse the gene is widely expressed at a low level. Immunolocalization demonstrated that the protein is cytosolic, with a higher concentration in the region of plasma membranes, especially apical membranes. The authors suggested that kanadaptin may be part of the machinery that targets kAE1 to the basolateral membrane. |

Application Note | Tested in peptide ELISA. WB: Use at an assay dependant dilution. Detects a band of approximately 130-150 kDa (predicted molecular weight: 92 kDa). Please note that currently we cannot find an explanation in the literature for the band we observe given the predicted size of approx 92KDa according to NP_060628. Current literature suggests RAT Kanadaptin to be approx. 53kDa (PMID: 9422766). We would appreciate any feedback from people in the field - have any results been reported with other antibodies/lysates? Have any splice variants/modified forms been reported? Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |
