Background | The INK4a ARF locus encodes two unrelated proteins both of which function in tumor suppression. p14 ARF arrests the cell cycle in a p53 dependent manner. p14 ARF binds to mdm2 and promotes the rapid degradation of mdm2 protein required for mdm2 modification and concurrent p53 stabilization and accumulation. This interaction is mediated by the exon 1 alpha encoded N terminal domain of p14 ARF and a C terminal region ofmdm2. Deletion of the ARF INK4a locus simultaneously impairs both the INK4a cyclinD/cdk4 RB and the ARF mdm2 p53 pathways. |

Application Note | Immunofluorescence: assay dependent. Immunoprecipitation (native): use at a concentration of 2?g / mg. Western Blotting: use at a concentration of 2 - 4?g / ml for 2hrs at RT. Not tested in other applications. Optimal dilutions / concentrations should be determined by the end user. By Western blot, this antibody detects a band of 14 kDa, which corresponds to the predicted molecular weight of p14 ARF. |
Specificity | p14ARF previously was known as p16 beta. This antibody reacts with 14kD p14ARF. |
