Background | ZAP70, a 70 kDa member of the Syk tyrosine kinase family, plays a central role in lymphocyte activation and development, and is implicated in several immune disorders. Upon T cell antigen receptor (TCR) engagement, ZAP70 is phosphorylated on tyrosines 292, 315 and 319 in the interdomain B, located between the SH2 and kinase domains. Phosphorylation of both tyrosines 315 (a Vav binding site) and 319 (a Lck binding site) enhances ZAP70 function in mediating lymphocyte signaling, while tyrosine 292 terminates the transient activation of ZAP70 and attentuates lymphocyte signaling. Phosphorylation of tyrosines 315 and 319 plays an important role in mediating the positive and negative selection of T cells in thymus. |

Host | Mouse |
Isotype | IgG2a |
Immunogen | Fusion to tandem SH2 domains of human ZAP70 corresponding to residues 1-254. |
Antigen Species | Human |
Tested Applications | IHC (Formalin-fixed paraffin-embedded sections). The usefulness of this product in other applications has not been determined. |
Application Note | This antibody may be diluted to a titer of 1:50-1:75 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. We suggest an incubation period of 60 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. |
