Background | GDP-dissociation inhibitors or GDIs, modulate the activation of GTPases, and may also be involved in the cellular compartmentalization of GTPases. GDIs modulate GTPase activity by binding to small G Proteins, and regulating GDP/GTP exchange. This family of proteins includes Rho-GDI (GDI-1), D4 GDI (GDI-2) and Rho-GDI-3 (GDI-3). Rho-GDI and D4 GDI are both localized in cystol. RhoGDI and GDI/D4 are both members of the Rho subfamily of GTPases. These two proteins are highly homologous, differing primarily in the first 25 amino acids of the N-terminus. D4 GDI has been described as a Caspase 3 (CPP32/YAMA/Apopain) substrate during Fas-induced apoptosis and is located in the cytosol. |

Application Note | IP: 4 ¥ìg is recommended to immunoprecipitate D4 GDI from 500 ¥ìg of Jurkat RIPA lysate. WB: Use at a concentration of 0.5 - 1 ¥ìg/ml. THis concentration is recommended for blotting using RIPA lysates from human A431, HL-60 and Jurkat cells, anti-rabbit IgG-peroxidase conjugate and a chemiluminescent detection system. Predicted molecular weight: 28 kDa. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. |
Specificity | Ab12427 specifically reacts with D4 GDI and does not recognize Rho-GDI or GDI-3. |
