In the electron microscope neurofilaments appears as 10nm diameter fibres of indeterminate length which generally have fine wispy protrusions from their sides. They are found particularly abundantly in axons of large projection neurons. They probably function to provide structural support for neurons and their synapses and to support the large axon diameters required for rapid conduction of impulses down axons. They are composed a mixture of subunits which usually include the three neurofilament triplet proteins, known as NF-L, NF-M and NF-H. Neurofilaments may also include smaller amounts of peripherin, a-internexin, nestin and in some cases vimentin. Antibodies to the various neurofilament subunits are very useful cell type markers since the proteins are among the most abundant of the nervous system, are expressed only in neurons, and are biochemically very stable. Species: Reactivity: | NB 300-222 reacts with NF-M from human, mouse, rat, cat and chicken. Other species have not been tested. | Applications: Uses: | NB 300-222 can be used in Western blot and immunofluorescence experiments. On blots NB 300-222 runs ~145-170kDa. Suggested working dilutions: * Western Blot - 1:10,000 or lower (ECL) Immunofluorescence - 1:2,000 *The investigator should determine the optimal working dilution for a specific application. | Dilutions: | immunofluorescence 1:2000, Western Blot 1:10000 | Positive Controls: | 1 positive controls | Packaging: Buffer: | PBS (10mM isotonic 0.9%, w/v), pH 7.2 | Storage: | Store at 4C short term. Aliquot and store at -20C long term. Avoid freeze-thaw cycles. | Preservative: | 10mM Sodium Azide | Limitations: | This product is for research use only and is not approved for use in humans or in clinical diagnosis.
